We have just published a paper in ASC Photonics, Optimizing Effective Labeling Efficiency in MINFLUX 3D DNA-PAINT Microscopy by Maximizing Marker Detection Probability.

Optimizing Effective Labeling Efficiency in MINFLUX 3D DNA-PAINT

The gist is that we noticed needing considerable imaging time to exhaustively image the markers in densely labeled samples. We tested this systematically and show that even for small areas 1-2h is a minimum. With standard DNA-PAINT markers a natural end to acquisition results from quite considerable site-loss in DNA-PAINT. We show that this can be overcome with the repeat domain sequences we introduced in 2021 for widefield SMLM in a Nature communications paper. With that approach the high labeling efficiencies obtained in widefield super-resolution can be attained in 3D MINFLUX imaging.
